An anti-pan Caveolin-1 antibody has been coated onto a 96-well plate. Samples are pipetted into the wells and Caveolin-1 present in a sample is bound to the wells by the immobilized antibody and the wells are washed. In select wells, mouse anti-phospho-Caveolin1 (Y14) antibody is added to detect phosphorylated Caveolin-1. In the remaining wells, rabbit anti-pan-Caveolin1 antibody is used to detect pan Caveolin-1. After washing away unbound antibody, HRP-conjugated anti-mouse IgG or HRP-conjugated anti-rabbit IgG is pipetted into the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Caveolin-1 (Y14) or pan Caveolin-1 bound.
