Closely associated is CRKL, or CRK-like protein (36 kD). CRKII and CRKL possess a C-terminal SH3 domain and a regulatory tyrosine phosphorylation site that are lacking in CRKI. CRK’s SH2 and SH3 domains serve as positive and negative regulators of transformation, respectively. CRK is vital to cell motility via activation of Rac1 by CRKII and CRKL and cell branching and adhesion via activation of Rap1 by CRKI.
