Autoimmune reactivity against DLAT is the most common cause of primary biliary cholangitis, an autoimmune disease caused by autoantibodies against mitochondrial antigens. Additionally, mutations in this gene can cause pyruvate dehydrogenase E2 deficiency, resulting in primary lactic acidosis in infancy and early childhood.
