The PH domain interacts with membrane phosphoinositides, while PRD domain interacts with SH3 domain-containing proteins. Dynamin-2 is a large mechanochemical GTPase involved in various cellular processes such as endocytosis, apoptosis, cytokinesis, phagocytosis, neuronal morphology, axonal growth, cell migration, cell membrane trafficking and cytoskeletal regulation. The studies indicate that mutations in Dynamin-2 cause centronuclear myopathy (CNM), Charcot-Marie-Tooth disease dominant intermediate type B (CMTDIB), hereditary spastic paraplegia and lethal congenital contractures syndrome type 5 (LCCS5).
