Its expression is highly controlled by the tumor suppressor protein p53, thus, p21 is involved in controlling the G1 cell cycle arrest in response to different stress stimuli. p21 plays a regulatory role in S phase DNA replication by interacting with PCNA (proliferating cell nuclear antigen). Activation of cyclin-dependent kinase 2 has been observed after p21 is cleaved by CASP3-like caspases. Loss of function of p21 has been associated with bladder cancer, Multiple Endocrine Neoplasia Type I and lymph node metastasis.
