FOXO1 binds to the conserved insulin-responsive elements and acts as a transcriptional activator. The FOXO1 target genes are involved in cell metabolism, growth, and differentiation. Upon insulin stimulation, FOXO1 proteins are phosphorylated by AKT\/PKB through the PI3K pathway, resulting in the translocation of FOXO1 proteins from the nucleus to the cytoplasm. The expression of FOXO1 target genes are then inhibited by FOXO1. FOXO1 has been reported to regulate maturation and class-switch recombination of B cells, as well as participating in regulatory T cell function.
