The enzyme hydrolyzes oxygen-phosphorus bonds in inorganic pyrophosphate and nitrogen-phosphorus bonds in phospholysine, phosphohistidine, and imidodiphosphate (2). LHPP expression is downregulated in multiple cancer types (3). The protein acts as a tumor suppressor, inhibiting proliferation, migration, and invasion and promoting apoptosis during cancer development (4), probably through its inhibitory effect on Akt and TGF-Î²\/SMAD pathway signaling (5-8). The gene has been linked to major depressive disorder (MDD), also known as clinical depression (9). In the lysosome, LHPP mediates inorganic pyrophosphate (PPi) hydrolysis (i.e., PPi to Pi) to facilitate vacuolar ATPase (V-ATPase)-dependent proton transport across the lysosomal membrane.
