Originally identified as a binding partner of JunD and ATF4, FAM175B stabilizes ATF4 by inhibiting its ubiquitin-dependent degradation, leading to activation of the ATF4-CHOP pathway and induction of apoptosis under endoplasmic reticulum stress.  In esophageal squamous cell carcinoma, FAM175B expression is down-regulated and acts as a p53-independent tumor suppressor; restoring its expression reduces proliferation and increases apoptosis of p53-mutated tumor cells.
