Lipocalin family members carry small hydrophobic ligands such as pheromones. However, the physiological functions of MUP1 remain poorly understood. Microarray and real-time PCR analyses revealed that MUP1 mRNA abundance in the livers of db\/db obese mice was reduced approximately 30-fold compared to their lean littermates. This change was partially reversed by treatment with the insulin-sensitizing drug rosiglitazone. In both dietary and genetic obese mice, circulating MUP1 concentrations were significantly lower than in lean controls. Chronic elevation of circulating MUP1 in db\/db mice using an osmotic pump-based protein delivery system increased energy expenditure and locomotor activity, raised core body temperature, and reduced glucose intolerance and insulin resistance. At the molecular level, MUP1-mediated improvements in metabolic profiles were accompanied by increased expression of genes involved in mitochondrial biogenesis, enhanced mitochondrial oxidative capacity, reduced triglyceride accumulation, and improved insulin-evoked Akt signaling in skeletal muscle (but not in the liver).
