(Human CD262 concentration is pretty low in normal plasma, it may not be detected in this assay).
This assay employs an antibody specific for Human CD262 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD262 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human CD262 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD262 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
We have not been able to detect endogenous Human CD262 in normal serum with ab113340, only in serum spiked with Human CD262.
Optimization may be required with urine samples.

Properties and Storage Information:
Shipped at conditions-Blue Ice, Appropriate short-term storage conditions--20°C, Appropriate long-term storage conditions--20°C, Storage information--20°C

Supplementary Information:
This supplementary information is collated from multiple sources and compiled automatically.
TRAIL R2 also known as Death Receptor 5 (DR5) is a type of receptor involved in the process of programmed cell death or apoptosis. The protein is approximately 55 kDa in size. TRAIL R2 belongs to the tumor necrosis factor receptor superfamily. It is expressed in many tissues including the spleen thymus and liver. TRAIL R2 interacts with the TNF-related apoptosis-inducing ligand (TRAIL) to initiate apoptosis by activating intracellular signaling cascades.
Biological function summary
TRAIL R2 functions by transmitting apoptotic signals from the cell surface to the intracellular environment. TRAIL R2-ligand binding leads to the formation of a death-inducing signaling complex (DISC). This complex also includes Fas-associated protein with death domain (FADD) and initiator caspases like caspase-8. Upon DISC formation multiple downstream effects activate executing caspases such as caspase-3 which leads to apoptosis of the targeted cell.
Pathways
TRAIL R2 participates in both the extrinsic apoptotic signaling pathway and the NF-kB signaling pathway. The extrinsic pathway relies on ligand-induced receptor trimerization involving proteins like FADD and caspase-8 leading to apoptosis. In both pathways NF-kB modulation by TRAIL R2 can result in either cell survival or apoptosis depending on different cellular contexts.
TRAIL R2 has significant implications in cancer and immune-related conditions. In cancer TRAIL R2 is notable as its expression may either promote tumor cell death or in some cases contribute to resistance against apoptosis. The protein is closely connected with TRAIL and other death receptors like DR4 which are often studied for therapeutic targeting in cancer treatment. Moreover imbalances in TRAIL R2 signaling have associations with autoimmune diseases where deregulated apoptosis may lead to unchecked immune cell survival and cause disease progression.
