Standards and samples are pipetted into the wells and DPPIV present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse DPPIV antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of DPPIV bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Optimization may be required with urine samples

Properties and Storage Information:
Shipped at conditions-Blue Ice, Appropriate short-term storage conditions--20°C, Appropriate long-term storage conditions--20°C, Storage information--20°C

Supplementary Information:
This supplementary information is collated from multiple sources and compiled automatically.
The enzyme Dipeptidyl Peptidase IV (DPPIV) also known as CD26 is a serine protease with an approximate mass of 110 kDa. It is expressed in various tissues throughout the body including the kidney liver and small intestine. DPPIV plays a significant role in cleaving dipeptides from the N-terminus of polypeptides which influences the activity of many bioactive peptides and proteins.
Biological function summary
DPPIV functions in the regulation of immune responses and glucose metabolism. It is not typically part of a complex structure operating independently to modulate peptide hormones and cytokines. This regulation can affect insulin regulation and gut hormones critical for maintaining energy homeostasis and immune balance in the body.
Pathways
DPPIV interacts with metabolic and immune pathways. It plays a central role in the glucose metabolism pathway affecting substrates like GLP-1 a hormone involved in insulin secretion. Moreover in the immune system DPPIV interacts with molecules such as adenosine deaminase influencing T cell activation and proliferation pathways.
DPPIV has a strong link with type 2 diabetes and cancer. In diabetes altered DPPIV function affects insulin secretion and glucose homeostasis. For cancer its enzymatic activity influences tumor progression and immune evasion. The interaction with proteins like adenosine deaminase also affects immune response modulation which can impact the development or progression of these diseases.
