It forms the icosahedral capsid by creating a trimeric complex that covers most of the viral surface. This protein is highly expressed in adenovirus-infected host cells during the late phase of infection. Hexon protein contains loops that present variable regions which are targets for the host immune response.
Biological function summary
The hexon protein contributes critically to the stability and structure of adenovirus particles and assists in viral entry and disassembly inside the host cell. As part of the adenovirus capsid complex it interacts with other viral proteins such as penton base and fiber proteins. This protein also plays a role in immune evasion by binding to cellular molecules that modulate host immune response.
Pathways
The hexon protein is involved in the viral entry pathway influencing the attachment and internalization of the virus into host cells. The internalization often involves interactions with the CAR (Coxsackie and adenovirus receptor) protein. Additionally it participates in the adenovirus replication pathway where multiple viral proteins including the DNA polymerase coordinate for successful viral replication within the host cell.
Adenovirus type 1 hexon E11 is associated with respiratory tract infections and conjunctivitis. These infections can occur particularly in immunocompromised individuals. The hexon protein interacts with host proteins including integrins to mediate infection of epithelial cells therefore contributing to the pathology of these conditions.
