Prepare lysosome staining solution by diluting the Lysosome Probe 1:500 in your buffer of choice. Aliquot to avoid freeze thaw cycles. Note: Probe concentration may need to be adjusted according to cell type and assay conditions. Optional: Include live-cell nuclear counterstain such as DRAQ5™ (Cat. No. 424101). Add an equal volume of Lysosome Staining solution to cell culture media. Incubate the cells at 37°C for 30 minutes. Wash cells by twice with PBS or HBSS. If cells are in suspension, spin down and resuspend in buffer of choice. Add desired volume of PBS or HBSS to cells for observation.
