PTEN is commonly mutated in human cancer, hence, loss of expression\/function of PTEN due to mutations have been observed in prostate cancer, glioblastoma, endometrial, lung and breast cancers. Other non-cancerous clinical disorders associated with PTEN mutation are Cowden syndrome, Bannayan-Riley-Ruvalcaba syndrome, and proteus-like syndrome. PTEN is phosphorylated at several residues and some lead to increase in activity while others lead to loss of function and degradation.
