MTHFD1 consists of two major domains, an N-terminal containing the dehydrogenase and cyclohydrolase activities and a larger synthetase domain in the C-terminus. According to reports, fibroblasts of patients with MTHFD deficiency exhibited enriched MTHFD1 in the nucleus, leading to low rate of de novo dTMP synthesis. Mutations in the MTHFD1 gene in pregnant women are associated with increased risk of giving birth to children with neural tube defects, along with possible decreased fetal survival.
