The mammalian IRAK molecular family consists of four members (IRAK1, IRAK2, IRAK3\/IRAK-M, and IRAK4). Two are active kinases, IRAK1 and IRAK4, and two are inactive kinases, IRAK2 and IRAK3. However, all are involved in the regulation of the nuclear factor-κB (NF-κB) and mitogen-activated protein kinase (MAPK) pathways. The binding of IL-1 to IL-1 receptor type I (IL-1RI) initiates the formation of a complex that includes IL-1R1, IL-1RAcP, MyD88, and IRAKs. Unlike its family members IRAK1 and IRAK4, which promote inflammatory responses, IRAK3 (IRAK-M) functions as a negative regulator, helping to maintain immune homeostasis and prevent excessive inflammation. It is predominantly expressed in monocytes, macrophages, and dendritic cells, where it plays an essential role in dampening TLR-mediated immune activation. IRAK3 prevents the dissociation of IRAK1 and IRAK4 from MyD88, thereby disrupting downstream activation of TRAF6. It also inhibits the phosphorylation and degradation of IRAK1, which is a crucial step in propagating TLR signaling. Moreover, IRAK3 suppresses NF-κB activation, reducing the transcription of inflammatory cytokines. This regulatory role is essential for preventing excessive inflammation and avoiding conditions such as sepsis and chronic inflammatory diseases.
