The four PDGF chains assemble into disulfide-bonded dimers via homo- or heterodimerization. Five different dimeric isoforms have been described so far; PDGF-AA, PDGF-AB, PDGF-BB, PDGF-CC, and PDGF-DD. PDGF is a major mitogen for fibroblasts, smooth muscle cells, and others. They exert their functions by interacting with two related receptor tyrosine kinases, PDGF receptor α (PDGFRα) and PDGF receptor  β (PDGFRβ). Upon ligand binding, PDGFRα undergoes dimerization and autophosphorylation, resulting in the formation of docking sites for SH2 domain-containing signaling proteins.
