GSK-3α contributes to insulin regulation of glycogen synthesis by phosphorylating and inhibiting glycogen synthase (GYS1 or GYS2) activity and mediates the development of insulin resistance. In Wnt signaling, GSK-3α regulates the level and transcriptional activity of nuclear CTNNB1\/beta-catenin. GSK-3α is also reported to facilitate the generation of amyloid precursor protein (APP)-derived amyloid plaques found in Alzheimer disease.
