Several NFATC3 variants generated by alternative splicing were identified. NFATC3 plays an important role in the regulation of T cell activation-related gene expression, especially in the induction of IL-2.  NFATC3 is expressed in immune cells, such as double positive (DP) thymocytes, mature CD4 + T cells, and mast cells. It is also found in non-immune cells such as smooth muscle and Schwann cells. In the resting T cells, NFATC3 is located in the cytoplasm and is phosphorylated. Upon T cell activation by TCR stimulation, the increase of intracellular Ca2+ induces NFATC3 dephosphorylation via calcineurin. Dephosphorylated NFATC3 exposes the nuclear localization signal and is subsequently translocated from the cytosol to the nucleus.
