The eIF3 complex directly links the 43S pre-initiation complex to eIF4F-bound mRNA. The subunit eIF3F has been shown to negatively regulate both cap-dependent and IRES-dependent translation and also interacts with mTOR and S6K1, and therefore also regulates mTORC1 and mTORC2 signaling. EIF3F mRNA is downregulated in multiple types of cancer. Overexpression of eIF3F protein leads to hypertrophy by altering protein synthesis via the mTORC1 pathway.
