This protein can act as an insulator protein by binding to an unmethylated imprinting control region (ICR) structure and prevent the activation of cis-acting elements which in turn prevents gene activation. CTCF becomes further involved in gene regulation when it forms a complex with cohesin. This complex can stabilize the formation of a chromatin loop, which can promote or repress the binding of other transcription factors to the promoter and enhancer, leading to gene activation or gene repression respectively. CTCF has a role as a tumor suppressor, when the gene goes through a mutation or deletion there is a rise in variance in CpG methylation throughout the genome.
