Channel activity seems to be regulated by a calmodulin-dependent mechanism with a negative feedback mechanism. Defects in TRPV4 are the cause of brachyolmia type 3 (BRAC3), spondylometaphyseal dysplasia Kozlowski type (SMDK) and metatropic dysplasia (MTD).
