Localized in the cilia, it is required for cilium biogenesis and sonic hedgehog signaling. Defects in ARL13B are the cause of Joubert syndrome (JS) which is an autosomal recessive disorder characterized by a distinctive cerebellar malformation (PMID: 19906870).
