LIRs are transmembrane proteins containing either two or four extracellular immunoglobulin domains, and have diverse functions, including the regulation of inflammation, immune tolerance, cell differentiation and nervous system plasticity (PMID: 16406677; 26040207). LILRB3, also known as ILT-5 or CD85a, belongs to the subfamily B class of LIR receptors which contain two or four extracellular immunoglobulin domains, a transmembrane domain, and two to four cytoplasmic immunoreceptor tyrosine-based inhibitory motifs (ITIMs). LILRB3 is found on the surface of a variety of cell types including monocytes\/macrophages, granulocytes, NK cells and some T cells. It binds to MHC class I molecules and transduces a negative signal that inhibits stimulation of an immune response.
