Cells were pelleted then lysed with Lysis Buffer (50 mM Tris-HCl pH 7.5, 150 mM NaCl, 10 mM NaF, 1x Protease Inhibitors, 1% SDS) and the lysate cleared by sonication. The protein concentration was measured by BCA assay. Modified 5x SDS sample buffer was added to the lysate before storage at -80°C.
