A monoclonal antibody from mouse specific for KLK12 has been pre-coated onto 96-well plates. Standards and test samples are added to the wells, a biotinylated detection polyclonal antibody from goat specific for KLK12 is added subsequently and then followed by washing with PBS or TBS buffer. Avidin-Biotin-Peroxidase Complex is added and unbound conjugates are washed away with PBS or TBS buffer. HRP substrate TMB is used to visualize HRP enzymatic reaction. TMB is catalyzed by HRP to produce a blue color product that changed into yellow after adding acidic stop solution. The density of yellow is proportional to the Human KLK12 amount of sample captured in plate.
Kallikrein-12 is a protein that in humans is encoded by the KLK12 gene. It belongs to the kallikrein subgroup of serine proteases, which have diverse physiologic functions in many tissues. KLK12 is expressed primarily in salivary gland, stomach, uterus, trachea, prostate, thymus, lung, colon, brain, breast, and thyroid gland and at lower levels in testis, pancreas, small intestine, and spinal cord. The expression of KLK12 is downregulated at the mRNA level in breast cancer tissues and is upregulated by steroid hormones in breast and prostate cancer cell lines.

Properties and Storage Information:
Shipped at conditions-Blue Ice, Appropriate short-term storage conditions--20°C, Appropriate long-term storage conditions--20°C, Storage information--20°C

Supplementary Information:
This supplementary information is collated from multiple sources and compiled automatically.
Kallikrein 12 also known as KLK12 belongs to the kallikrein family of serine proteases. This family plays significant roles in various physiological processes. Kallikrein 12 has a mass of approximately 27 kDa. It is expressed in a range of tissues but shows higher expression in the skin respiratory tract and brain tissues. This distribution suggests that Kallikrein 12 may have diverse functional roles depending on its location in the body.
Biological function summary
The KLK12 protein influences complex processes involving extracellular matrix remodeling and possibly tissue homeostasis. It functions as part of a larger group of kallikreins which collectively impact the proteolytic cascade mechanisms. The activity of KLK12 is regulated by specific inhibitors which maintain balance in protease activity within the tissues ensuring normal cell and tissue functions.
Pathways
KLK12 plays a role in protease cascades involved in tissue repair and inflammation. The kallikrein-kinin system is one of the critical pathways where KLK12 contributes. This system affects blood pressure regulation and inflammatory responses. KLK12 associates with proteins such as kininogens and other kallikreins which together facilitate its involvement in these biological processes impacting both vascular biology and immune responses.
KLK12 has associations with certain skin disorders and respiratory diseases. Alterations in KLK12 expression are observed in conditions like asthma where the protein's role in inflammation may influence disease progression. KLK12 associates with other kallikrein family members in these disorders highlighting the potential for multiple kallikreins to impact pathophysiological conditions. Improper regulation of KLK12 activity can contribute to these disease states making it a potential target for therapeutic interventions.
