When used correctly, various shades of pink can be obtained to aid in visualization of tissue components. Erythrocytes, collagen, and the cytoplasm of muscle or epithelial cells will stain with different shads of pink.\u003cbr\u003e\nCytoplasm: Pink to Red.\u003cbr\u003e\nErythrocytes Pink to Red.\u003cbr\u003e\nNuclei: Black\/Blue (Hematoxylin).\u003cbr\u003e\nConterstain:\u003cbr\u003e\nAny fixed or frozen tissue. Cell smear.\u003cbr\u003e\nProcedure (Standard):\u003cbr\u003e\n1. Deparaffinize sections if necessary and hydrate to distilled water.\u003cbr\u003e\n2. If sections are Zenker-fixed, remove mercuric chloride crystals using iodine and clear with sodium thiosulfate. Rinse in running tap water.\u003cbr\u003e\n3. Rinse slide in distilled water.\u003cbr\u003e\n4. Stain slide in Hematoxylin, Mayer's for 5 minutes.\u003cbr\u003e\n5. Rinse slide in running tap water for 2-3 minutes.\u003cbr\u003e\n6. Apply Bluing Reagent for 30 seconds.\u003cbr\u003e\n7. Rinse in distilled water.\u003cbr\u003e\n8. Stain slide in Eosin Y Solution for 5 minutes.\u003cbr\u003e\n9. Rinse quickly in 95% alcohol followed by 2 minutes in absolute alcohol.\u003cbr\u003e\n10.
