CASP4 is expressed as an inactive proenzyme comprised of a pro-domain and a large and small protease subunit. CASP4 is activated by cleavage of an internal Aspartic residue, thus generates a heterodimeric enzyme consisting of the two subunits which is capable of cleaving and activating its own precursor protein. Overexpression of CASP4 results in apoptosis.
