Hemopexin is the plasma protein with the highest binding affinity for heme. When oxidized, hemoglobin that circulates alone in the plasma (also known as free hemoglobin) turns into methemoglobin. Methemoglobin then further dissociates into free heme alongside the globin chain. When further oxidization occurs, the free heme will turn into free met-heme. This will cause the hemopexin to bind to the free met-heme, forming a met-heme and hemopexin complex until it reaches a receptor on hepatocytes or macrophages within the spleen, liver, and bone marrow. Evidence suggests that brain and blood iron homeostasis is perturbed in Alzheimer’s diseases.
