SMYD2 localizes to the nucleus and cytoplasm and methylates histones such as H2B, H3K4, H3K36, and H4 as well as non-histone substrates, notably p53 on lysine 370. As a protein lysine methyltransferase, SMYD2 is a regulator of transcription and epigenetic signaling and functions in the regulation of cell cycle and apoptosis. It has also been shown to promote NF-κB signaling pathways via methylation of TRAF2 and STAT3. Aberrant SMYD2 expression is associated with a variety of cancers and other diseases including kidney disease, cardiovascular disease, and rheumatoid arthritis.
