PDHA1 and PDHB form the heteroterameric pyruvate dehydrogenase E1 (E1) subunit of the PDC. The enzymatic activity of the PDC is regulated by phosphorylation of PDHA1 through pyruvate dehydrogenase kinase (PDK) and pyruvate dehydrogenase phosphatases (PDP). PDCs play crucial physiological functions between fed and fasting states which regulate pyruvate metabolism to maintain glucose homeostasis. PDHA1 enzymatic dysfunction causes fatal lactic acidosis and chronic neurologic dysfunction in the central nervous system. The decreased expression of PDHA1 in Parkinson’s disease and dementia with Lewy bodies patients suggests a dysfunction of mitochondrial energy production.
