In hematopoiesis, HOXB4 enhances the self-renewal capacity of hematopoietic stem cells (HSCs), which are responsible for the continuous production of blood cells. Overexpression of HOXB4 in HSCs has been shown to lead to an expansion of these stem cells without inducing leukemogenesis. By binding to specific DNA sequences, HOXB4 activates a network of target genes that promote stem cell proliferation and inhibit differentiation, thus maintaining the stem cell pool.  HOXB4 also plays a pivotal role in organogenesis, particularly in the development of the spinal cord and the patterning of the vertebral column. During embryonic development, HOXB4 is expressed in a specific spatial and temporal manner, contributing to the formation of structures along the anterior-posterior axis. Its expression helps determine the identity and positional information of various segments, ensuring that body parts develop in the correct locations. In vertebrates, this function is crucial for the proper segmentation and alignment of the spine, as well as for the formation of the nervous system.
