Selection and loading of peptides onto MHC I is orchestrated by the peptide-loading complex (PLC). Tapasin, a central component of the PLC, stabilizes MHC I and catalyzes the exchange of low-affinity against high-affinity, immunodominant peptides. In this quality-control step, tapasin cooperates with other more general, ER-resident chaperones, including ERp57 and TAP. Tapasin has been found to be downregulated in glioblastoma, renal cell carcinoma, prostate cancer, urothelial carcinoma, cervical carcinoma, head and neck carcinoma and esophageal squamous cell carcinoma.
