Background Fatty acid desaturase 1 (FADS1), also known as delta-5 desaturase, is essential for the biosynthesis of highly unsaturated fatty acids (HUFAs) from linoleic acid (LA) and α-linolenic acid (ALA), both of which are polyunsaturated fatty acids (PUFAs). In addition, FADS1 converts NADH to NAD . Studies show that FADS1 and fatty acid desaturase 2 (FADS2) thus enable NAD recycling for glycolysis to support cell viability when aerobic respiration is acutely suppressed (1,2). Furthermore, FADS1 and ELOVL5 expression is enhanced in mesenchymal-type gastric cancer cells, sensitizing the cancer cells to ferroptosis. On the contrary, FADS1 and ELOVL5 expression is inhibited in intestinal-type gastric cancer cells, leading to resistance to ferroptosis (3).
