Background N-acetyl-D-glucosamine kinase (NAGK) is a member of the sugar kinase/Hsp70/actin superfamily, whose members phosphorylate certain substrates in an ATP-dependent manner (1-3). NAGK mediates the phosphorylation of N-acetylglucosamine (GlcNAc) to GlcNAc-6-phosphate, which is then used for biosynthesis of UDP-GlcNAc, a critical component of O-linked-N-acetylglucosaminylation and N-linked glycosylation (1-4). NAGK is expressed mainly in immune cells and is co-expressed with genes primarily associated with immune cell functions (4). NAGK can also phosphorylate muramyl dipeptide (MDP), triggering pro-inflammatory gene expression by NOD2 (nucleotide-binding oligomerization domain-containing protein), which is critical for maintaining a healthy intestinal barrier function (4).
