ThPOK is best known as a signature CD4+ T cell transcription factor that is upregulated during the differentiation of CD4+ T but not CD8+ cytotoxic T cells (1). Expression of ThPOK in developing T cells represses expression of CD8 and cytotoxic T cell effector genes, and indirectly promotes expression of CD4 by antagonizing RUNX-mediated CD4 repression (2-4).
