Background Rab32 is a small GTPase best known for its role in regulating membrane trafficking, including endosomal pathway regulation and Golgi apparatus structure and localization (1,2). Rab32 is an essential component of a host defense pathway for defending against intracellular pathogens, helping to link lysosomes and bacteria-containing phagosomes (3,4). Rab32 also recruits IRG1, an enzyme that produces the antimicrobial metabolite itaconate, to pathogen-containing vacuoles (5). Altered Rab32 function is implicated in the pathogenesis of Parkinson's disease. The Ser71Arg mutation, which results in increased activation of LRRK2 and reduced colocalization of Rab32 and PINK1, is associated with development of Parkinson's disease (6).
