Background XRCC3 (X-ray repair cross-complementing protein 3) is one of five RAD51 paralogs that form two distinct complexes, BCDX2 (RAD51B, RAD51C, RAD51D, and XRCC2) and CX3 (RAD51C and XRCC3) (1). Along with RAD51, these complexes are crucial in the repair of DNA double-strand breaks through homologous recombination (2), and in the response to DNA replication stress (3,4). The CX3 complex also plays a role in maintaining mitochondrial DNA integrity during replicative stress (5). XRCC3 genetic polymorphisms are associated with risk and prognosis in human cancer (6-9).
