SerpinB1 blocks the activity of caspase-1, caspase-4, and caspase-5; the protein does so via suppression of caspase recruitment domain oligomerization and enzymatic activation (1). SerpinB1 is expressed in neutrophils and monocytes, and the protein resides in the cytoplasm due to its lack of a secretory signal sequence (2,3). There, SerpinB1 protects cells from serine proteases that may leak from granules into the cytoplasm, including Cathepsin G, neutrophil elastase, proteinase-3, and Granzyme H (4,5).
