Product Usage Information For optimal ChIP results, use 10 μL of antibody and 10 μg of chromatin (approximately 4 × 10 6 cells) per IP. This antibody has been validated using SimpleChIP ® Enzymatic Chromatin IP Kits. Western Blotting: 1:1000 Simple Western™: 1:50 - 1:250 Immunoprecipitation: 1:100 Chromatin IP: 1:50 Storage Supplied in 10 mM sodium HEPES (pH 7.5), 150 mM NaCl, 100 µg/mL BSA, 50% glycerol, and less than 0.02% sodium azide. Store at -20°C. Do not aliquot the antibody. Background POZ/BTB and AT hook containing zinc finger 1 (PATZ1) is a transcription factor that regulates gene expression activation or repression depending on the cellular context (1-3). The human gene has four alternatively spliced variants that give rise to structurally similar isoforms. PATZ1 protein contains a BTB domain that facilitates protein interaction and AT hook and zinc finger domains that enable its DNA binding (4,5). PATZ1 has been shown to play key regulatory roles in embryonic development and stem cell identity, proliferation, senescence, and apoptosis (6-10). It has also been reported that PATZ1 plays a critical role in cancer cells, where it appears to carry out both oncogenic and anti-tumor activities (8-10). Notably, PATZ1 is downregulated in thyroid cancer and, thus, thought to function as a tumor suppressor in these cells (11).
