Background Lymphocyte antigen 6 family member G6D (LY6G6D) is a member of the lymphocyte antigen 6 (Ly-6) superfamily that is located within the MHC class III region of chromosome 6 (1). LY6G6D is anchored to the cell membrane by glycosylphosphatidylinositol (GPI) and is believed to play a role in filopodia formation, but its exact function is not fully understood (2,3). LY6G6D is overexpressed in colorectal cancer tumors but has minimal expression in healthy tissue, making it an attractive target for immunotherapies (4). Indeed, bispecific T cell engagers targeting LY6G6D and CD3 have shown initial promise in clearing colorectal tumors in mice (5,6).
