SPARC proteins (secreted protein acidic and rich in cysteine) are secreted glycoproteins involved in tissue renewal and remodeling, and embryonic development (1). SPARC proteins are highly upregulated in melanoma and other tumor types, but their contributing functions are not yet understood (2). However, the role SPARC proteins play in regulation of extracellular matrix interactions is likely to contribute to melanoma development and metastasis. SPARCL1 is likely to play a similar role in extracellular matrix remodeling in the vasculature and the brain. For example, SPARCL1 is upregulated in vascular endothelial cells in response to injury and infection, both remodeling the extracellular matrix and promoting pro-inflammatory macrophage activation (3).
