It senses calcium levels in neurons and is primarily located in the outer segments of photoreceptors within the vertebrate retina, where it participates in light adaptation via calcium-dependent inhibition of the enzyme rhodopsin kinase (RK) (1). Rcv, like other family members, contains four EF-hand motifs and a myristoyl chain on the N-terminus. Only two of the EF-hands (EF2 and EF3) bind Ca across species (1). In darkness, Ca -bound Rcv inhibits G protein-coupled receptors (GPCRs) and thereby prevents visual pigment phosphorylation. Under bright light conditions, Ca dissociates from Rcv and the protein translocates to the inner segment. By this, Rcv prolongs phototransduction and enhances visual sensitivity (3).
