MPP8 forms a dimer that serves as a bridge between G9a, GLP, and DNMT3A to set up cytosine methylation and H3K9 tri-methylation at target loci (4). MPP8 is a member of the human silencing hub (HUSH) complex along with TASOR, PPHLN1, and MORC2, which localizes to H3K9me3 sites and cooperates with SETDB1 to maintain H3K9me3 levels and transcriptional repression (5). The HUSH complex is capable of silencing long interspersed elements (LINEs) located in euchromatic regions (6).
