ATF7IP has a SUMO binding domain, which interacts with sumoylated MBD1 to recruit the SETDB1\/ATF7IP heterodimer to sites of heterochromatin (4). Loss of ATF7IP results in degradation of SETDB1 protein and a global loss of H3K9me3 (5). ATF7IP can localize to PML bodies in a SUMO-dependent manner, causing reduced histone mRNA and protein and cellular senescence (6). ATF7IP has also been implicated in cancer, where it interacts with SP1 to regulate telomerase.
