It is a key enzyme in ketogenesis (1,2). Studies show that HMGCL expression increases in BRAF V600E-expressing human melanoma cells, resulting in upregulated levels of intracellular acetoacetate. Acetoacetate enhances binding of BRAF V600E to MEK1, stimulating activation of the MEK-ERK signaling pathway. Suppression of HMGCL leads to reduced proliferation of these human melanoma cells (2).
