Chitinases are part of the 18-glycoside hydrolase (18-GH) family of proteins that act to bind and degrade chitin. In mammals, this function provides defense against chitin-containing pathogens. Chitotriosidase-1, also known as Chitinase-1 or CHIT1, is the most abundant of these enzymes. CHIT1 consists of an N-terminal catalytic domain, a C-terminal chitin-binding domain, and a short hinge region that connects the two. CHIT1 activity is implicated in several inflammatory signaling pathways and is considered a sensitive biomarker for activated immune cells. It is produced and secreted by macrophages, microglia, and neutrophils in normal and inflammatory conditions. Elevated levels of CHIT1 in serum is a suggested biomarker for Gaucher's disease, atherosclerosis, pulmonary fibrosis, and some cancers.
