It promotes axon growth by activating the mitogen-activated protein kinase (MAPK) pathway through Î²1 integrin receptors. When expressed on activated T cells, it promotes the production of proinflammatory cytokines by macrophages through the Î±1Î²1 integrin receptor (1). Semaphorin-7A may also play a role in resolving inflammation through its regulation of macrophage chemotaxis and chemokinesis, and by promoting macrophage differentiation and polarization toward an anti-inflammatory phenotype (2). Neuroinflammation is a detrimental component of neurodegenerative diseases, such as Parkinson's disease (PD) and multiple sclerosis (MS). The role that semaphorin-7A plays in mediating inflammatory responses in microglia in the brain suggests that it may be a therapeutic target for these diseases.
