FBXL11 maintains heterochromatin and promotes epigenetic silencing by associating with centromeres and recruiting HP1 (3). Furthermore, FBXL11 is recruited to non-methylated CpG islands by its zinc finger CxxC domain (3). FBXL11 can also control ribosome biogenesis by preventing the transcription of rRNA genes (4). During DNA damage, FBXL11 is phosphorylated by ATM, which reduces its ability to bind to chromatin, allowing H3K36 dimethylation to accumulate and recruit the MRE11 complex (5). Additionally, there is a short isoform of FBXL11 that lacks a JmjC domain and histone demethylation activity.
