Zinc finger protein 366 (ZNF366) was originally identified in a yeast two-hybrid screen for transcriptional regulators responding to estrogen (3,4). ZNF366 (dendritic cell (DC)-specific transcript, DC-SCRIPT) is a member of the Cys-Cys:His-His subfamily of zinc finger proteins (5). ZNF366 localizes to the nucleus and does not show transcriptional activation via transactivation assays but does interact with co-repressors (2,5). ZNF366 has been shown to be DC-specific and is expressed in myeloid DCs, plasmacytoid DCs, and Langerhans cells (5). ZNF366 knockout mice show an immune deficiency phenotype, with substantially decreased expression of the transcription factor, IRF-8 (6). Loss of function in ZNF366 has been implicated in tumor cell evasion of immune responses (7).
